DSIP (Delta Sleep-Inducing Peptide) is an endogenous neuropeptide studied for its roles in sleep regulation, stress response, and neuroendocrine function.

DSIP, explained

The short version: DSIP (delta sleep-inducing peptide) is a nine-amino-acid peptide isolated from rabbit blood in 1977 and named for the deep sleep it seemed to trigger. When a 2006 review called it a riddle, its gene and receptor were still unknown, and the sleep link remains weak.

What it is

A group in Basel isolated DSIP in 1977 from the venous blood draining the brains of rabbits, and it was first seen as a candidate natural sleep factor. Its sequence does not match any known peptide family. Strangely for a hormone-like molecule, no gene, precursor protein or receptor for it has been identified.

What has been studied

By the mid-1980s a review in Peptides could list a long run of animal studies on sleep and on effects beyond sleep, with researchers exploring it for insomnia, pain and withdrawal. The mechanism was still unexplained.

In people, a 1992 double-blind study in Neuropsychobiology gave DSIP or placebo to 16 people with chronic insomnia over three nights. Sleep efficiency was a little higher and falling asleep a little quicker on DSIP, but the effects were weak, partly down to a chance change in the placebo group, and subjective sleep quality did not budge. The authors concluded it was unlikely to be of major benefit for chronic insomnia.

Where the evidence stops

A 2006 review in the Journal of Neurochemistry called DSIP a still unresolved riddle and described the case for it as a sleep factor as extremely poorly documented. The Moscow authors noted that in their own rabbit and rat work, some synthetic DSIP analogues promoted deep sleep but DSIP itself did not. The human work we found is small and dates from the 1980s and early 1990s.

Handling and storage

Named for an effect it never convincingly showed. An interesting puzzle, not an established sleep peptide.
