5-Amino-1MQ is a small molecule inhibitor of NNMT that may increase intracellular NAD+ levels by blocking a key catabolic pathway. It offers a distinct route to NAD+ elevation compared to NMN or NR precursors.

The short version: 5-Amino-1MQ is a small molecule, not a peptide, that blocks an enzyme called NNMT. Almost everything known about it comes from mice and cells, largely from one university lab in Texas.
The full name is 5-amino-1-methylquinolinium. It inhibits nicotinamide N-methyltransferase (NNMT), an enzyme that puts a methyl tag on nicotinamide, a building block of NAD+, and uses up SAM, the body's main methyl donor, while doing it. In fat cells, blocking NNMT with this compound lowered the enzyme's end product and raised both NAD+ and SAM. That is the link to NAD+ that our journal piece on the two digs into.
The target came first. A 2014 Harvard study in Nature found NNMT levels were higher in the fat and liver of obese and diabetic mice, and that switching the gene down in those tissues protected mice from diet-induced obesity by raising energy use.
A team at the University of Texas Medical Branch then built drug-like inhibitors and named 5-amino-1MQ as its lead (Biochemical Pharmacology, 2018). In diet-induced obese mice treated for 11 days, treated mice lost about 5% of body weight while controls gained about 1.4%. A white fat depot shrank by about 35% and fat cells got more than 30% smaller, with no difference in how much the mice ate.
The same lab's NNMT inhibitor was tested in 24-month-old mice with an injured leg muscle (2019). Muscle stem cells were more active, fibres grew back larger, and peak muscle force was about 70% higher than in untreated old mice.
