AOD-9604 is a modified fragment of human growth hormone (hGH172-191) that may retain the fat-metabolising properties of GH without activating the full GH receptor, making it useful for isolated lipolysis research.

The short version: AOD-9604 is a modified piece of the tail end of human growth hormone, developed in Australia as an obesity drug. It went through six human trials, looked safe, and did not deliver the weight loss it was built for.
AOD-9604 is a 16-amino-acid peptide. It takes positions 177 to 191 of human growth hormone, adds an extra tyrosine at the front to make it more stable, and closes a ring between its two cysteines. It grew out of Monash University work on the part of growth hormone that seemed to drive fat breakdown, and Metabolic Pharmaceuticals took it into clinical development.
It is close kin to HGH Fragment 176-191 but not the same thing. AOD-9604 is one specific, modified molecule with its own safety studies and human trials. The plain fragment never had either.
In obese rodents, the Monash group reported less weight gain and more fat breakdown in fat tissue. Toxicology followed in rats and cynomolgus monkeys.
Then came people. A 2013 review pooled six randomised, placebo-controlled trials. AOD-9604 did not raise IGF-1, did not worsen glucose tolerance the way full growth hormone can, triggered no antibodies in the people tested, and no serious adverse events were put down to it. The problem was efficacy: weight loss seen in earlier trials did not show up in the last, larger trial, which included an intensive diet and exercise programme. It never became an obesity medicine. It later gained GRAS (generally recognised as safe) status for use in foods and supplements, on condition that the safety data were published.
The human trials asked one question, whether it causes fat loss, and the final answer was no better than placebo. Claims beyond that rest on rodent and tissue work. WADA names AOD-9604 on its Prohibited List as a growth hormone fragment. The trial safety record belongs to the trial material, not to research vials.
